Do a search here - I've posted Vegas to dos a lot. I also suggest utilizing Yelp. There are a ton of Vegas Yelpers with some great "must-do" Vegas lists that you can see on that site.

Off Off-Strip:
-Red Rock Canyon: Gorgeous drive. Hiking, etc. if you want it. Also puts you close to Red Rock Casino, Bonnie Springs, etc.
-Hoover Dam: Walk across the new bridge. Do the full dam tour (it is the most expensive one, but the only one that actually takes you inside the actual dam). Stand in two places at once (NV and AZ).
-Boulder City: Right by Hoover Dam. Only city in all of NV with absolutely no gaming. A good brewery. Tons of antiques.
-Bootleg Canyon: Right by Hoover Dam and Boulder City. Has a fantastic ziplining experience.
-Valley of Fire: It's Red Rock Canyon on steroids. One of the most beautiful things you will ever see.
-Area 51: Tiny town of Rachel, NV. ET highway. Infamous black mailbox. It's awesome.
-Nevada Test Site: Must reserve well in advance and submit SS#, etc. Coolest experience ever.

Off-Strip:
-Neon Museum: #1 must-do recommendation in Vegas. You get to see all the old neon signs and hear all of the history behind them. And you get to take awesome pictures. Must reserve tour in advance. Near downtown area / Cashman Field.
-Atomic Testing Museum: So few people know this part of Vegas's history. It's part of the Smithsonian. Extremely well-done. Very close to the Strip (not walkable per se, but super short cab ride).
-Fremont Street: Fremont Street Experience and East Fremont. Giant canopy of lights, restored neon signs, cheap gaming, Jesus protesters, awesome bars (Griffin, DCR, etc.), awesome hangouts (The Arts Factory), Street Eats (second Saturdays), ziplining, etc. And this is the original Strip and where all that is Vegas began.
-Pinball Hall of Fame: Not far off the Strip on Tropicana. No charge, just quarters to play the games (most are $0.50). So many really old pinball machines. And so neat to read the little handwritten histories of each of them.
-Hard Rock Casino: Tons of rock memorabilia all throughout the casino areas.
-Las Vegas Premium Outlets: High-end outdoor outlet mall. Near downtown, right by the Clark County government center.

On-Strip:
-Mob Experience: Inside the Tropicana. Pretty neat interactive experience thing. It's still really new and they are still working out some of the kinks.
-Stratosphere: Go to the top twice - once at night and once during the day. Skip the lines and paying and all that bullshit by going to the Level 107 Lounge.
-CityCenter: Download the CityCenter Art app - use it as a guided walking tour of all of the cool art.
-Eiffel Tower: Go to the top day or night and watch the Bellagio fountain show.
-Big Elvis: Best free show on the Strip. Performs 3x daily at Bill's Gamblin' Hall.
-All the other free shows, particularly the Lake of Dreams at the Wynn (sit at Parasol Down and get a cocktail and watch it) and the Living Garden at the Palazzo.
-Just walk through the various casinos, particularly the Cosmopolitan, Paris, Mirage, Aria, Venetian, Wynn/Encore, and Caesars Forum Shops.
-Eat all of the amazing restaurants (please don't eat at any of the chains or fast food places, it's just sad).

Not worth your money:
-The Secret Garden: Saddest little "zoo" ever.
-Shark Reef: Neat, but small, tons of kids running around, and so not worth the price.
-Dolphin Habitat: Not worth the price. And sometimes you won't actually even get to see anything more than one dolphin swimming under water.

It may not be warm enough, but the pools are open now (most of them anyways - some don't open until mid-April). So there are also all of the day parties at the different pools and whatnot. And I definitely recommend spending some time at a spa.

Personally, for first timers especially, I suggest heading to the top of the Stratosphere (during the day) so you can get a 360 view of the Las Vegas Valley and see how big it is, the mountains that surround it, etc. Few people realize how big Vegas is, that it is in fact in a valley, etc. And I cannot HIGHLY recommend enough the Neon Museum / Boneyard. It's amazing to see, but it'll also give you a good rundown of Las Vegas history (which I find very fascinating). http://www.neonmuseum.org/tours.html
